Localization of 1,25-dihydroxyvitamin D/sub 3/ in intestinal nuclei in vivo

Autoradiography of frozen sections of intestinal tissue taken from rachitic chickens given a single intravenous dose of 1,25-dihydroxy(23,24/sup 3/H)vitamin D/sub 3/ (650 pmol, 78 Ci/mmol) has been carried out. Specific localization of label in the nuclei of intestinal villi and crypt cells could be demonstrated at 2.5 to 6 h postintection. In contrast, no concentration or localization of radioactivity could be detected in intestinal muscle, liver, and skeletal muscle. These results strongly support the concept that the function of 1,25-dihydroxyvitamin D/sub 3/ in intestine is mediated by a nuclear mechanism.
