Polyamines and flowering in thin layer tobacco tissue cultures. III. Kinetics of uptake and binding of labelled spermidine
/sup 14/C-labelled spermidine (Spd) was supplied to thin layer tobacco tissue cultures growing on Murashige-Skoog medium + 1 ..mu..M NAA and kinetin. Such cultures produce only flower buds, which become obvious after about 15 days. Spd is taken up rapidly at a rate sustained throughout the culture period. Considerable binding of Spd to TCA - precipitable material occurs by day 2 of culture; it increases to a maximum on day 4 and declines by day 10, shortly before flowering. About 80% of the TCA - precipitable label remains in the form of Spd, 12% shows up as putrescine and 3% as spermine. Differential centrifugation revealed binding of Spd to various cell organelles. DNase, RNase and proteases were used to liberate Spd from the bound form. In nuclei it appears that 60% of the label is associated with nucleic acids and 40% with the protein fraction. In view of evidence from animals indicating Spd binding to specific proteins during organogenesis, they are investigating the nature of the Spd binding protein(s).
