Nephrotoxic effect of cadmium as cadmium chloride or cadmium metallothionein in renal cortical slices

The mechanism of cadmium-metallothinoein (CdMT) induced nephrotoxicity was studied in isolated rat kidney cortical slices. Accumulation of the organic anion, p-aminohippuric acid (PAH) and the organic cation, tetraethyl ammonium (TEA) into renal slices (0.5 mm) was measured after in vitro exposure of the slices with different concentrations of Cd/sup 2 +/ (5 x 10/sup -6/ to 2 x 10/sup -4/) as either CdCl/sub 2/ or CdMT for 90 minutes at 25/sup 0/C under 100% O/sub 2/. The concentrations of Cd, PAH and TEA were measured in the slices and media after incubation. The uptake of TEA was not significantly altered in the presence of either CdCl/sub 2/ or CdMT while PAH uptake by the slices was inhibited by both Cd compounds. The inhibitory effect of PAH uptake was dose dependent with both Cd compounds, however CdMT caused a greater effect at low doses as compared to CdCl/sub 2/. This may be due to a preferential uptake of Cd as CdMT by the slices. The intracellular presence of zinc metallothionein (ZnMT) or the in vitro addition of zinc salts may affect the toxic effect of these Cd compounds. The results show that the inhibitory effects of the Cd compounds were specific for the anionic transport in renal slices.
