Toxicity of D- and L-selenomethionine in Fisher 344 rats

Weanling female Fisher 344 rats were fed ad libitum, a commercial control diet to which was added selenium (Se) as sodium selenite, 16 ppm Se; D- or L-selenomethionine at 10 or 16 ppm Se. Animals (15/diet) were weighted, sacrificed and major organs were weighed, fixed in formalin and frozen for Se analysis. Compared to controls, mean body weights of animals fed 16 ppm Se as selenite, D- or L-selenomethionine were significantly reduced. L-selenomethionine produced the greatest weight reduction at 12 wks of age. D- and L-selenomethionine at 10 ppm Se did not significantly reduce body weight compared to controls. Se toxicity as measured by change in organ weights and histological examination were only observed in animals receiving 16 ppm Se diets. Analysis of liver and muscle at 12 wks of age showed significantly higher Se deposits in rats fed D- or L-selenomethionine than selenite. Slight variations were observed in fecal Se. Large differences in D- and L-selenomethionine toxicities were not observed, but L-selenomethionine appeared to be slightly more toxic. Toxicity was not overt in animals fed 10 ppm Se. Toxicity from Se as selenite was most severe.
