Does tris(o-phenanthroline)ruthenium(II) intercalate in DNA? The definitive answer from scanning force microscopy

Determining the mode-of-binding of polypyridyl metal complexes to DNA has proven to be problematic. Despite considerable efforts by a number of research groups, the binding of even the most well-studied compound of this group, tris(o-phenanthroline) ruthenium(II) (Ru(phen){sub 3}{sup 2+}), is still under debate. We have recently established scanning force microscopy (SFM) as an assay for intercalation. Lengthening of individual DNA molecules caused by intercalation is directly visualized by SFM and serves as direct evidence for intercalative mode-of-binding. We have assayed the binding of both enantiomers of Ru(phen){sub 3}{sup 2+} to DNA using SFM. Since no lengthening of the DNA is observed, we conclude that both the {Delta} and {Lambda} enantiomers bind purely by non-intercalative modes.
