Identification of acetate groups on the backbone of rhamnogalacturonan I, a pectic polysaccharide of primary walls, obtained from suspension cultured cotton cells

Pectic polymers are an integral part of the primary cell walls of dicots. Pectins may play a role in cell expansion, cell adhesion, disease responses, and perhaps cell differentiation and recognition. The structure of pectins is complicated and is not fully characterized. A more detailed knowledge of their structure will allow a better understanding of the function of pectins. The disaccharide GalA-Rha which is the repeating unit of the backbone of the polymer was isolated by treating the cell walls with anhydrous liquid HF at -23 C for 30 minutes. Liquid HF under these conditions extracts all of the sugar from the walls except those in cellulose and the homogalacturonan region of pectins while retaining the acyl substituents on the sugars. This treatment takes advantage of the enormous difference in lability in HF of rhamnosyl and galacturonosyl linkages. /sup 1/H NMR spectra of the disaccharide show that around one in 2 of the repeat units is acetylated. Preliminary results from methylation analysis indicate that the acetate group is most likely on the 2 or 3 position of the galacturonic acid.
