Radiation potentiating effect of ethyl bis(2,2-dimethyl-l-aziridinyl) phosphinate (AB-163)

Ethyl bis(2,2-dimethyl-l-aziridinyl) phosphinate (AB-163), a new tris(aziridinyl) phosphine oxide (TEPA) analogue, was tested in a murine P388 lymphocytic leukemia combination radiotherapy chemotherapy model. In this system, the combination was synergistic with results rivaling the best previously reported radiation drug combination results in this model. Three time intervals between the two modalities were tested. The optimum combination utilized AB-163 120 mg/kg four hours before 400 rad whole-body ..gamma..-radiation; this resulted in 50% survival at 100 days and 330% increased life span (ILS) for the remainder. Drug alone produced a maximum of 188% ILS (120 mg/kg) with one survivor while radiation alone gave 48% ILS (optimum 400 rad) with no survivors. These findings make AB-163 attractive for further investigation including clinical combined modality trials.
