Effects of gamma irradiation on sensitivity of antigen presenting cells (APC) to transforming growth factor beta (TGF-B)

Spleen cells, freshly isolated Langerhans cell containing epidermal cells (fEC/LC), and 72 h cultured epidermal cells (cEC/LC) were either irradiated and incubated 3 h at 37C, or irradiated immediately before addition to MLR. When the {minus}3 h protocol was used, both fEC/LC and spleen exhibited a 50% reduction in allostimulatory capacity compared to 0 h irradiated cells. Conversely, {minus}3 h irradiated cEC/LC stimulated a 2 fold greater response than 0 h cEC/LC. Cultured EC/LC, subjected to the different irradiation protocols, were compared for susceptibility to inhibition of allostimulatory capacity by TGF-B. Cells which were irradiated before TGF-B treatment stimulated only 50% of the response of non-similar responses whether pretreated with TGF-B or not. Spleen cells treated with TGF-B and then irradiated were profoundly sensitive to TGF-B, whereas similarly treated cells subjected to mitomycin C in place of irradiation were TGF-B resistant. These data suggest that APC which appear capable of antigen presentation immediately after irradiation may actually have radiation induced lesions rendering them sensitive to the effects of treatment with cytokines. These findings emphasize the importance of standardization of irradiation protocols in in vitro functional studies on the effects of cytokines on cells of the immune system.
