The effect of zinc deficiency on erythrocyte (RBC) deformability as measured by ektacytometer

Experiments were performed to investigate the influence of Zn deficiency on RBC deformability. Weanling, male Wistar rats were fed diets containing < 1 ppm Zn ad libitum for 3 weeks. Controls were fed ad libitum or pair-fed a diet containing 100 ppm Zn. Time zero controls were also included. Blood was sampled from the 4 groups, and RBC deformability was measured at standard shear stress under conditions of increasing osmolality in the ektacytometer. Elongation index was plotted against osmolality. When characteristics of the osmotic spectrum were analyzed by a one-factor ANOVA, maximum cell deformability and Omax were found to be increased between 3 and 6 weeks of age in the rat. The increase in Omax suggests an increase in the water content of the cells. The drop in voluntary food intake which accompanies zinc deficiency significantly decreased EImaz. When the zinc-deficient and pair-fed groups were compared by a paired t-test, the zinc deficient group also showed significantly depressed EImaz. The decrease in EImax is due to either an increase in membrane surface area or a change in membrane shear modulus.
