THE ROLE OF ADRENAL CORTICAL HORMONES IN X RAY-INDUCED LIVER CHANGES

Total-body irradiation of rats with 600 r produced severe degenerative lesions in liver which were evident by the 6th or 7th day. A dose of 400 r had no visible effects on liver and 500 r produced slight changes in some animals, seen as fat accumulation at the periphery of liver lobules. At the 600-r dose the following histopathologic changes were seen in liver: separation of trabeculae and dilatation of sinusoids in central portions of lobules, severe fatty infiltration at the center of lobules, and desquamation of sinusoid endothelial cells. In adrenalectomized rats, maintained on deoxycorticosterone acetate (DOCA), 600 r failed to produce any liver lesions in -- half of the animals and fatty degeneration was mild in the remainder. In these animals, irradiation appeared to have no effect on the spleen and lymph nodes, which underwent hypotrophy as a result of adrenalectomy. In adrenalectomized rats treated with 1.5 mg/day hydrocortisone in addition to DOCA, irradiation produced the same liver lesions as in irradiated controls. Thus, glucocorticoid hormone appears to sensitize the liver to radiation injury. (H.H.D.)
