Title: EFFECTS OF TOTAL IRRADIATION OF THE RAT ON THE FREE ACTIVITY OF ACID DEOXYRIBONUCLEASE AND OF ACID PHOSPHATASE IN THE REGENERATING LIVER (in French)

Journal Article · · Arch. intern. physiol. et biochim.

Rats were partially hepatectomized and irradiated with 700 r at various subsequent intervals. Homogenates were prepared of the regenerating liver, and in the supernatant fraction, obtained by centrifugation at 300,000 g for 30 min, determinations were made of the activity of acid-deoxyribonuclease (DNase), liberated into this fraction, and of another enzyme present in the lysozomes, acid phosphatase. Irradiation did not affect the free acid-DNase in nonhepatectomized rats but when given 2 hr before hepatetomy, it doubled this activity as tested 4 hr later. Irradiation given immediately after hepatectomy also doubled this activity by the 6th hr, but the activity returned to normal by the 18th hr. When the interval between hepatectomy and irradiation was prolonged (up to 24 hr), irradiation was decreasingly effective in releasing free enzyme. Closely similar results were obtained with acid phosphatase as with acid-DNase. The rise in activity of free enzymes refiects a lesion in the lysosomes produced by radiation; apparently the lysosomes are most radiosensitive in the early stages of regeneration of liver.
