White, female mice 2.5 to 3 months in age were given a single 200 r dose of x rays on the 12th to 19th day of pregnancy Male members of the litter were killed on attaining an age of 45 days, and testicles were weighed and subjected to histological analysis. Transverse slices of the seminal canals (100) were selected, and a quantitative estimate was made of the different types of embryonic cells: spermatogonia, prespermatocytes, spermatocytes, spermatids, and sperms. The percent of barren canals in the testicles was also used as a quantitative indicator of the amount of damage to the testicle. The maximum decrease in weight of the testicle, which serves as a good indicator of the degree of damage caused by the radiation, was observed for irradiation on the 16th day of embryogenesis. The weight of the testicle of a 45 day old animal was only 20 mg as compared to a testicle weight of 125 mg in the controls, The histological results showed that the fetus undergoes extensive damage with irradiation on the 15th to 18th days of embryonic development. Maximum damage was observed for irradiation on the 16th day of embryogenesis. Active spermatogenesis was taking place in all the canals of the testicles of the 45 day control mice. No barren canals were observed in the controls The percent barren canals was 0.14% for irradiation on the 12th day of pregnancy. There was a gradual increase in the percent barren canals for irradiation on the 13th, 14th, and 15th days with a maximum of 96.1% on the 16th day of embryogenesis. Then, the percent barren cells gradually decreased to 6.8% barren canals on the 19th day of embryonic development. It was proved that a dose of 200 r causes severe damage to the gonads, especially when the irradiation is performed on the 14th to 17th day of embryogenesis. (TTT)
