PURIFICATION AND BIOLOGICAL CONTROL OF A PREPARATION OF INSULIN LABELED WITH IODINE-131

S>An evaluation was made of the denaturation, degradation, and biological inactivation of insulin accompanying its labeling with I/sup 131/, and modifications in the labeling procedure are described. These modifications entail removal of I not bound to protein and separation of insulin-I/sup 131/ from degraded protein. Inorganic and organic nonprotein I can be removed by dialysis or chromatography on Amberlite resin. in the 1st procedure the product contained 16 plus or minus 3% I and in the 2nd, 3 to 12%. Separation of insulin-I/sup 131/ from degraded protein was achieved by paper chromatography or cellulose column chromatography. Paper chromatography revealed 3 radioactive fractions, the intermediate peak (R/sub f/ 0.2) corresponding to that of native insulin and accounting for 66% of the total radioactivity. Biological tests on insulin-I/sup 131/ (in vivo in dogs and in vitro on rat epididymal fat) showed that 20 to 70% of its physiological activity is lost by iodination. The described technic yields a preparation with 0.65 atoms I per molecule insulin with a specific activity of 37 mu c/unit. (H.H.D.)
