EARLY CHANGES OF THE GROWTH ZONE IN THE RABBIT FOLLOWING ROENTGEN IRRADIATION: AUTO-RADIOGRAPHIC INVESTIGATION AFTER THE ADMINISTRATION OF RADIOSULPHATE

Histologic changes and uptake of S/sup 35/O/sub 4/ by chondrocytes were studied in the epiphysial cartilage of young rabbits irradiated locally with 1000 r. Damage was perceptible in osteoblasts in metaphysis 6 hr after treatment. The osteoblasts were partially necrotic with pyknotic nuclei and were dislodged from the bone spicules. Blood-forming cells were reduced in number, and these changes persisted. No changes were seen in the epiphysial cartilage itself 6 or 48 hr after treatment, but 4 days after irradiation, abnormally large portions of the cartilage were seen to be devoid of cartilage cells. Pronounced changes in the epiphysial cartilage were present 10 days after irradiation. Proliferating cartilage cells were then concentrated in irregular aggregations separated by defibrillated matrix. The bone trabeculae on the iaradiated side were shorter and fewer than normal. No deflnite changes in S/sup 35/uptake were perceptible 6 or 48 hr after irradiation, whether the S/sup 35/ had been given 2 hr, 24 hr, or 5 days before death. Four days after irradiation, uptake of S/sup 35/ 2 hr after the injection was less pronounced on the irradiated side, and some areas in the cartilage zone showed no activity. Ten days after irradiation the clumped cartilage cells were labeled, but their uptake differed from normal. Thus, 2 hr after S/sup 35/ injection some cells in the proliferative zone were active in their entirety, while a number of large cells in the hypertrophic zone showed only slight activity. Thus, roentgen irradiation has a pronounced effect on growth, although this is not definitely demonstrable until 10 days later. (H.H.D.)
