DENATURATION OF DEOXYRIBONUCLEIC ACID OF ESCHERICHIA COLI BY X-IRRADIATION IN VITRO

Desoxyribonucleic acid (DNA) extracted from Escherichia coli was examined for the effects of x radiation on the ultraviolet absorption spectrum. Typical ultraviolet absorption curves obtained from nonirradiated, irradiated, and acid-denatured DNA solutions are presented. A comparison of the curves shows that the denaturation factor increased rapidly with dose, to about 541% of the maximum possible denaturation abserved in the acid denatured control. It is concluded that there is a progressive increase tn the disruption of hydrogen bonds as determined from the increase in the extinction coefficient with increasing radiation dose. (C.H.)
