THE PHARMACOLOGY OF RADIOPROTECTANT CHEMICALS. BIOCHEMICAL CHANGES IN THE DOG FOLLOWING THE ADMINISTRATION OF BETA-MERCAPTOETHYLAMINE (MEA)

General pharmacological reactions produced by slow intravenous injection of a large dose (100 mg/kg) of MEA in the dog are described. Epinephrine and norepinephrine levels in plasma increased during the early normotensive period and maximum levels were observed with profound hypotension, which occurred 9 to 15 min after the beginning of the administration of MEA. Elevated catechol amine levels persisted for 1 to 2 hr and gradually returned toward control values at the end of 3 hr. Diphenhydramine (10 mg/kg, intravenously, 30 min prior to the administration of MEA) increased the circulating levels of catechol amines; however, it suppressed the expected increase following MEA. Vagatomy also suppressed the expected levels of the catechol amines following MEA administration. Blood glucose increases occurred but later than the catechol amine elevations, and peak values were attained at approximates 1 hr after MEA injection. Diphenhydramine and surgical procedures, which suppressed MEA-induced increases in epinephrine, also tended to suppress blood glucose rises, and barbiturate anesthesia suppressed the blood glucose increases usually produced by MEA. Hypoxia was present as early as 10 min after the beginning of MEA injection, during the normotensive period. Oxygen capacity of the blood increased slightly, and this was associated with an increase in hematocrit values. Most the effects of MEA can be attributed to refease of epinephrine and norepinephrine from the adrenal medulla and the sympathetic nervous system. (P.C.H.)
