ON HEPATIC DYE EXCRETION WITH ALTERED BROMSULPHTHALEIN METABOLISM AFTER ROENTGEN IRRADIATION
The conjugation reaction of Bromsulfalein (BSP) with glutathione and excretion in bile as BSP mercaptide (and to a lesser extent with cysteine and glycine) was studied in irradiated animals. Rats were exposed to 1200-r wholebody doses of 180-kv x rays delivered at a rate of 60 r/min. At various subsequent intervals (6-12, 24, and 48 hr) they were injected intravenously with 10 mg/kg BSP, and excretion of the dye in bile was followed for 1 hr, at 10-min intervals. The total 60-min biliary excretion of the dye was not significantly affected 6 to 48 hr postirradiation. The values were 83, 91, and 84% (of the total amount injected) af 6 to 12, 24, and 48 hr postirradiation, compared with 83% in nonirradiated control rats. However, the conjugation rate was significantly depressed in irradiated animals, the 60-min values being 81, 81, and 70% at 6 to 12, 24, and 48 hr postirradiation, respectively, on the basis of 100% for control rats. Chromatographic studies showed that the BSP-glutathione conjugate was diminished in bile of irradiated rats but that the cysteine and glycine fractions were not affected. Experiments with liver homogenates of irradiated rats, to which reduced glutathione was added, showed a 20% decrease in ability to form BSP mercaptide. It was concluded that the enzyme system promoting conjugation is injured by radiation. However, the possibility that the transport and excretion mechanisms for free and conjugated BSP are also injured cannot be eliminated. (TCO)
