Ehrlich ascites cells incubated at 0 deg C incorporated much less orotic acid than those incubated at 37 deg . The percentage of labeled cells rose only over the first hour, and further incubation was unable to increase the precursor's incorporation. This indicates that at low temperatures penetration of orotic acid into cells is independent of metabolic activity. The rapid numerical increase of labeled cells and of the grains over nucleus and cytoplasm noted in the autoradiogram on incubation at 37 deg seems to be due to metabolic activity and RNA synthesis. On treatment with ribonuclease, radioactivity vanished in most of the cells, which suggests that nearly all the orotic acid was incorporated in RNA. Irradiation with 2300 r clearly depressed the percentage of labeled cells and modified the distribution of radioactivity as revealed by the localization of the grains. The number of cells with grains over nucleus and cytoplasm was larger after irradiation. It appeared that x rays had in some way promoted the precursor's penetration into the cells. On further incubation of unirradiated preparations, the number of cells with radioactivity in both the cytoplasm and nucleus increased rapidiy. All nonirradiated cells were labeled after 2-hr incubation, whereas in irradiated preparations an increase occurred only in the percentage of cells, with the precursor either only in nucleus or oniy in cytoplasm.
