RENAL TUBULAR FINE STRUCTURE STUDIED DURING REACTION TO ACUTE URANIUM INJURY

Acute U injury was studied in rats injected subcutaneously with 14.4 mg/ kg UO/sub 2/(NO/sub 3/)/sub 2/, as the hexahydrate salt. For the following 6 days urinary volume and protein content were measured, and renal tubules were examined by light and electron microscopy. Enlargement of subbasilar compartments, apical swelling, and apical vacuolation were conspicuous and maximal in the first 2 days after injury, at the time polyuria was developing. The changes appeared to be related to disturbed fluid transport across cells. Enlargement of subbasilar compartments occurred in proximal, distal, and collecting tubuies, implying that all 3 segments were concerned in fluid transport. Cytoplasmic vacuoles and bodies, including myelin figures and hyaline droplets, increased in number, size, and variety as proteinuria deveioped and necrotic debris appeared in tubular lumens. Necrosis, most extensive in proximal convoiuted tubules, was not identified prior to pyknosis by any of several earlier changes in the fine structure. Earlier changes included loss of microvilli of brush borders, swelling of mitochondria and endoplasmic reticulum and Golgi systems, focal to general cytoplasmic edema, and margination of nucleoplasm.
