Determination of Fe 55 and Fe 59 Mixtures in Blood by Liquid Scintillation Counting

Iron-55 and Fe59 were determined simultaneously in blood samples after the organic material had been destroyed by nitric and perchloric acids. The Fe was precipitated as hydroxide, dissolved in oxalic acid and electroprecipitated on a platinum rod. It was then dissolved as perchlorate, the solution evaporated to a small volume and added to a solution of diphenyloxazole in toluene and absolute ethanol. The activity was determined in a single photomultiplier tube liquid scintillation counter by means of pulse height analysis. The accuracy of the method (standard deviation) was 3.4%. Slightly more than 1.3% of the error was the combined standard error due to countering statistics, and the remaining 2% due to chemical, technical, and instrumental errors. The possible influence of different amounts of total Fe in the sample was also examined, because quenching by Fe might have caused a spectrum shift. In samples containing 2.5 to 20.0 mg stable Fe in addition to identical amounts of Fe55 and Fe59, no evidence of such an effect on the results was detected.
