AN EVALUATION OF FOUR CHEMICAL MUTAGENS FOR EFFECTIVENESS IN PRODUCING CHROMOSOMAL ABERRATIONS IN MOUSE LIVER. Final Report

Four radiomimetric substanoes were tested for the production of chromosomal aberrations in mouse liver. The LD-56 was determined first for each compound. The experimental mice received either a single intraperitoneal injection of this LD-50 or twice weekly injections of 25% of this dose. Controls received injections twice weekly of either saline or saline with Tween 80. The actual full doses and LD values obtained were: ethyl methane sulphonate 0.4 mg/gm (LD-10); Nitromin 0.12 mg/gm (LD-50); Myleran 0.036 rag/gm and chlorambucil 0.028 mg/gm (LD42); and dimethyl Myleran 0.02 mg/gm (LD-32). Five mice in each LD-50 group, 25% LD-50 group, saline and saline with Tween 80 groups were sacrificed monthly for four months after injections of CCl/sub 4/ to induce mitoses in the liver. Counts of chromosomal aberrations in 100 anaphases and telophases were made. When viewed individually, none of the compounds effected a great increase in chromosomal aberrations over those in controls; when viewed together however, all the compounds produced twice the number of aberrations over controls by the end of the first month, but this early decided effect was obscured eventually by the gradual increase in abnormalities with age in normal control mice. Many of the mice which had been injectad biweekly for some time with Myleran and chlorambucil or with dimethyl Myleran failed to respond to the regenerative stimulus of CCl/sub 4/, an effect indicative of the presence of a heightened mitotic rate in these livers. Control mice showed higher numbers of abnormalities than mice in previous experiments had shown. These results are discussed in terms of the somatic mutation theory of aging. (auth)
