A study was made of the effect of x rays and native, probably biologically active, deoxyribonucleic acid (DNA) prepared from the autotrophic culture of E. coli, upon the frequency of back-mutation in the auxotrophic thymine and uracil-requiring mutant (E. coli 15 T/sup -/U/sup -/). It was found that x rays can produce reversion in this mutant-strain, and autotrophic revertants (E. coli 15 T/sup +/U/sup +/) can be isolated after such treatment. The frequency of appearance of the revertants considerably increases if the mutant cultures are treated with native (highly-polymerized) isologous DNA prepared from autotrophic bacteria (E. coli 15) immediately after irradiation. This effect seems to be specific, because it has not been observed after treating the irradiated mutants with homologous and heterologous DNA, yeast RNA, and Bacto- peptone (Difco). The possible mechanism(s) of the effect of DNA on the process(s) of back mutation (reversion) induced by irradiation, is discussed. (auth)
