EFFECT OF WHOLE-BODY X IRRADIATION ON INDUCTION OF ALLERGIC ENCEPHALOMYELITIS IN RATS

The results indicate that 400-r whole-body x irradiation given to rats prior to sensitization with spinal cord and adjuvant suppresses allergic encephalomyelitis (AE). This suppression is dose-dependent and may be observed in two strains of rats sensitized by either of two routes. Conversely, whole- body x irradiation given after sensitization does not suppress AE. The data indicate that leukopenia does not account for suppression of AE in x-irradiated animals. Suppression of AE by x irradiation is most likely due to interference with immune responses to spinal cord plus adjuvant. Direct support for this view is provided by the fact that suppression of AE is paralleled by reduced complement-fixing anti-brain antibody formation. Doses of x irradiation used in this study appeared to exert no appreciable influence on development of delayed- type reactivity, as judged by tuberculin skin tests. The skin reactions to tuberculin observed in rats represented delayedtype immune responses against tuberculin antigens rather than heightened nonspecific cutaneous reactivity, e.g., brought about by prior injection of adjuvant. It is noteworthy that unequivocal positive tuberculin reactions could be elicited in x-irradiated rats in which AE was clearly suppressed. Results of the present study in the rat are consistent with the thesis that circulating anti-brain antibody may play a role in production of AE. (H.H.D.)
