Title: Ultraviolet mutagenesis as studied using the single-stranded-DNA phage OX- 174 (in Russian)

From international conference on the bases of the biological effects of ultraviolet radiation; Brno, Czechoslovakia (2 Oct Using the mutant am 3oX-174 phage containing single-stranded DNA as a model the reversion to the wild type induced by the action of uv light on the phage and the host bacterium was studied. The reversion frrquency under the action of uv light on the phage or the bacterium separately was shown to increase by 1.3 to 2 times only. The uv action on both the bacterium E. coli C (hcr/sup +/) or E. coli C/sub syn/ -(hcr/sup -/) and the phage caused the increase in the reversion frequency by 20 to 70 times as compared with the natural background (up mutagenesis). The uv mutagenesis degree was strongly dependent on the uv dose on bacterium and had a maximum value at 2OO to 300 erg/mm/sup 2/ for E. coli C and 20-30 erg/mm/sup 2/ for E. coli C/sub syn/- . Decrease in the concentration of pyrimidine dimers in the host DNA in the process of the photoreactivation caused a shift of the optimum of uv dose for phage reversion: at irradiation of the bacteria E. coli C--from 200 to 800 erg/mm/ sup 2/, at irradiation of the bacteria E. coli C/sub syn/- from 20 to 70 erg/mm/ sup 2/. The results show that genes controlling excision repair in bacteria (uvr or hcr) do not seem to be required for the phenomena of uv reactivation and uv mutagenesis in bacteriophage with single-stranded DNA.
