New radioimmunoassay technique using second antibody to solid phase applied to assays of human chorionic somatomammotropin

A modification of the double antibody method for the radioimmunoassay of Human Chorionic-Somatomammotropln (HCS) is proposed. Rabbit antibodies raised against goat gamma globulin are coated on polystyrene tubes under suitable conditions, to separate the anti-HCS - HCS complex from the test mixture. Thus the specific antibody and the sample to be tested, or the standard hormone are placed successively in the coated tubes. After incubation at 37 deg C for four hours, the labeled hormone is added. This is followed by an addltional incubation at 37 deg C for fourteen hours. The precision and sensitivity of the method, and the good agreement with results obtained by the usual double antibody system, indicate that the Double Antibody-Solid Phase Radioimmunoassay (DASP-RIA) is a valuable improvement in the clinical assay of HCS, and suitable for wider application.
