Tritium labeling of polyribonucleotides (in German)

Polyribonucleotides were labeled with tritium during incubation in tritium-contained water by an exchange reaction. A specific activity between 1 and 16 mli/mMol was obtained with pure polyribonucleotides: with increasing reaction time the polynucleotides decompose, the bases remain chemically unchanged. The chain shortening of the pure polyribonucleotide cannot be traced back to the BETA -radiation of tritium. Iodized cytidine diphosphate was chiefly labelled in position 5 with a specific activity of 30 mli/ mMol. In the enzymatic polymerization of 3-H-CDP to 3-H-poly(i), an isotope effect occurs that lowers the specific activity of the polymer by up to 20%. (GE)
