Studies on lysosomes after irradiation. I. A quantitative histochemical method for the study of lysosomal membrane permeability and acid phosphatase activity

A simple histochemical method was developed which enables the permeability of lysosomal membranes to be studied semiquantitatively and the relative specific activity of lysosomal acid phosphatase to be measured quantitatively in tissue sections after irradiation. Increase in membrane permeability, but not of enzyme activity, occurs after treatment of tissue sections with solutions of low pH or injection of vitamin A into the animal. Enzyme activity measured by the method has been related to standard determinations of tissue homogenates. (auth)
