Synchronization of the cell cycle of L-cells in the G$sub 2$-phase by fractionated x-ray irradiation and administration of Daunomycin. Quantitative determination of the degree of synchronization by an impulse cytophotometer (in German)

The synchronization effect of x rays and Daunomycin on an exponentially growing culture of L-929-cells was quantitatively tested with the impulse cytophotometer (ICP). By fractionated irradiation with 200-kV x rays (total dose 225 rad, 9 fractions of 25 rad each in intervals of 3 hrs), about 70% of the cells can reversibly be kept in the G/sub 2/-phase of the interphase. The portion of G/sub 2/-cells in the unirradiated culture amounts to l2.9%. Daunomycin likewise produces reversible G/sub 2/-blockage on the L-cell culture. After application of Daunomycin in the concentration of 0.05 mu g/ml, which proved to be optimal, after a 42 hr lasting period in influence, more than 80% of the cells were in the G/sub 2/-phase. An additivity of the synchronization effect of irradiation and Daunomycin was not observed. The results indicate that Daunomycin in the applied concentration also produces a G/sub 1//S-blockage that possibly is the cause for the lack of additivity of Daunomycin and x ray irradiation in the synchronization effect. The authors point to a potential application of the described synchrorization method in radiotherapy. (auth)
