Single-strand interruptions in newly synthesized DNA in Chinese hamster cells after x irradiation

DNA synthesized in cells of Chinese hamster V79/4 after x irradiation has segments of lower molecular weight than nonirradiated cells, as shown by pulse-labeling with tritiated thymidine, followed by sedimentation of the DNA in alkaline sucrose gradients. Postincubation of the x-irradiated cells increases the single-strand molecular weight of the newly synthesized DNA to the control level. With the same technique this was also shown by us after uv irradiation of Chinese hamster cells as it was earlier determined by others for mouse L 5178 Y cells, human cells, and also Chinese hamster cells. Possibilities are discussed to explain the decrease of single-strand molecular weight after x irradiation with 500 rad. (auth)
