Double radio-isotope derivative technique for the assay of drugs in biological material. I. The determination of maprotiline
The double radioisotope technique is described in general, and a sensitive and specific method is presented for the assay of maprotiline (Ciba 34 276-Ba, Ludiomil) in biological materials. The procedure involves the addition of a definite amount of /sup 14/Clabelled maprotiline as internal standard, extraction of the free base, and acetylation with /sup 3/H-labelled acetic anhydride. The /sup 3/Hlabelled N-acetyl derivative of maprotiline is then separated by two-dimensional thin-layer chromatography from /sup 3/H-active byproducts which arise from reagents and biogenic material. Both /sup 3/H- and / sup 14/C-activities of the purified acetyl derivative are determined simultaneously by liquid scintillation counting. The yield of /sup 14/C-activity makes it possible to evaluate the theoretical 100% /sup 3/H-activity of the derivative. A convenient method is described to determine the specific /sup 3/H- activity of the N-acetyl derivative. (5 tables) (auth)
