Radiosensitivity of resting and proliferating mouse-liver cells for mitotic delay and the inhibition of DNA synthesis

Groups of 75 mice were given 450 rads whole-body 250 kVp x rays either 4 hr before, 12 hr after, or 32 hr alter partial hepatectomy i.e., when most of the liver cells were in the G/sub o/ phase, G phase, and S phase respectively. All groups showed a delay in the onset of division and a reduction in total mitotic activity. Similar groups of 75 were given the same radiation dose at the same timepoints after surgery, then killed in groups of 15, 1 hr before which they were given intraperitioneally 1 mu Ci g/sup -1/ body-weight of tritiated thymidine. All irradiated groups showed a marked delay in the onset of /sup 3/H- labeling. noue having occurred 36 hr after operation. The irradiated groups also showed a marked reduction in the total number of cells undergoing DNA synthesis, with the exception of cells irradiated when they were regenerating and in DNA synthesis i.e., about 32 hr after partial hepatectomy.
