ON THE METABOLISM OF $sup 14$C LABELLED SEROTONIN IN VIVO

The metabolism of serotonin-C/sup 14/, labeled in the alpha position of the amino group, was studied in rats. In each of four experiments the serotonin was injected subcutaneously, and the animal was placed in a glass metabolism cage for quantitative collection of expired CO/sub 2/, urine, and feces. A blood sample was taken after 24 hours. The results showed that only a small fraction of the injected radioactivity was expired. The most rapid excretion of labeled CO/sub 2/ occurred 8 to 14 hours aiter serotonin administration. The preponderant fraction of the radioactivity was eliminated in the urine. The excretion was quite rapid, with 40% of the radioactivity being eliminated within the first twelve hours. Less than 2% of the original dose was eliminated through the feces. The distribution of the radioactivity in the organs and tissues was studied, and no activity was found in the brain. Small amounts were found in all other organs, with the spleen having the highest specific activity. Chromatographic studies of the urine showed that about 50% of the urine activity is caused by 5-hydroxyindoleacetic acid. (J.S.R.)
