SOUND-INDUCED SEIZURES IN LONG-SURVIVING PRENATALLY X-IRRADIATED RATS

Rats exposed to 250-r x radiation in utero, on either the 18th or 19th day of pregnancy, were tested for their susceptibility to sound-induced seizures at a median age of 21 1/2 months. No significant association between incidence of seizures and prenatal x irradiation was found. Some indication that x irradiation in utero enhanced the reactivity of seizure-susceptible animals was noted. It was concluded that audiogenic seizure susceptibility cannot be considered either a long-term or a sensitive indicator of the effect of x irradiation upon the central nervous system. The results are in conflict with those of Werboff, who reported an increase in seizure responses in the offspring of animals exposed to x radiation in the third trimester (day 15 and 20) of pregnancy, and tested at four months of age. The increase was unrelated to dosage (100 r, 50 r, or 25 r) or to day of gestation when x irradiated. They suggested that susceptibility to audiogenic seizure is a sensitive and long- lasting indicator of the effects of x irradiation upon the central nervous system. More generally, they concluded that their levels of x irradiation altered the threshold of susceptibility to sound-induced seizures when seizure susceptibility appears to be, at least in part, an age-related phenomenon, repeated testing of prenatally x-irradiated rats might demonstrate at specific periods of life, a significant relation between the early treatment e sound. Such an effect also presumably would be related to the dose of x irradiation to which the animalss were exposed in utero, and to the day of gestation when they were irradiated. (BBB)
