RESTITUTION OF LIVER MITOCHONDRIA IN WHOLE-BODY IRRADIATED MICE

Characteristics of swelling, the degree of coupling of oxidative phosphorylation-respiratory control, and P/O ratios of mitochondria in early post- irradiation stages were examined. White male mice were given a dose of 700 r x rays and killed 30 minutes, 1 hour, and 4 hours after exposure. Spontaneous swelling of liver mitochondria 30 minutes and 1 hour after exposure is markedly increased: an essentially normal condition if found at 4 hours. Contrasting of these results with those from thyroxine or Ca/sup 2+/-induced swelling, it is suggested that a slight alteration in structural integrity is involved in the early post-irradiation stage. The respiratory control index of mitochondria falls off 30 min after exposure and reaches a minimum 1 hour post-irradiation. A slight depression of P/O ratios appears only 1 hour after irradiation. Nearly complete or complete recovery of both respiratory control index and P/O ratio is achieved by 4 hours. Parallel with the biochemical lesion are the decrease and subsequent increase in the protein content of the mitochondrial suspension per gram of fresh liver tissue. From the results of this study and the electronmicroscopic observations of Mochituk, it appears that the mitochondria of mouse liver are transiently damaged both structurally and functionally in vivo, but are soon restituted to reach pre-irradiation levels.
