Organization of the deoxyribonucleic acid replicating system in mammalian cells as revealed by the use of x-radiation and bromuracil deoxyriboside

Results are reported from studies of the effects of x radiation on the synthesis of desoxyribonucleic acid in HeLa and hamster cells in tissue culture. The incorporation of tritium-labeled thymidine and bromouracil desoxyriboside, an analog of thymidine, was allowed for various time periods before or after irradiation at various doses. Three different methods of DNA extraction were used. Possible mechanisms for radioinduced inhibition of DNA synthesis are discussed. The concept of a very-high molecular weight DNA is discussed in relation to the data obtained.
