EFFECT OF NITROGEN MUSTARD POISONING AND TOTAL-BODY X-RAY IRRADIATION ON THE GLYCOGEN LEVEL OF THE LIVER

Rats were irradiated with 800 r, a dose equal to the LD/sub 50/30/. A second group of animals was treated with nitrogen mustard in a dose equivalent to LD/sub 50/30/. In formalinfixed livers periodic acid-Schiff (PAS) reaction showed the glycogen to have disappeared in 24 to 48 hr. Restitution sometimes took 30 days. Simultaneously a series of alcohol-fixed livers was studied histochemically. The glycogen content was seen to decrease in 48 hr with subsequent restitution generally taking 240 hr. The biochemical changes paralleled the histochemical pattern in alcoholfixed livers. To explain these observations it is suggested that the glycogen molecules in the liver, when exposed to the effect of radiation or nitrogen mustard, become less polymerized and thus more water-soluble. Proof of this was furnished by ultracentrifugal molecular weight determination of the glycogen extracted from the livers of the irradiated or nitrogen mustard-poisoned animals. When the liver glycogen specimens were grouped according to the degree of PAS positivity, and the samples with identical glycogen content were pooled and subjected to ultracentrifugation, the results supported this hypothesis.
