SENSITIVITY TO ULTRAVIOLET LIGHT OF SINGLE- AND DOUBLE-STRANDED DNA

The ultraviolet sensitivity of ss-DNA and rf-DNA were studied using spheroplasts of E. coli K12S hcr/sup +/ and hcr/sup -/ strains, respectively. No difference in uv sensitivity was observed when ss-DNA was assayed either on hcr/ sup +/ or on hcr/sup -/ spheroplasts. On the other hand uv-treated rf-DNA was much less biologically active when tested on hcr/sup -/ spheroplasts than on hcr/ sup +/ spheroplasts: its apparent uv sensitivity on hcr/sup +/ spheroplasts is approximately 1/8 that of ss-DNA whereas on hcr /sup -/ spheroplasts the uv sensitivity of rf-DNA is approximately equal to that of ss-DNA. The results indicate that in the absence of host cell reactivation ss- and rf-DNA show equal uv sensitivity and hence that rfand ss-DNA molecules incur an equal number of lethal hits at a given dose of uv light. Therefore, the difference in uv sensitivity between ss- and rf-DNA on hcr/sup +/ spheroplasts must then be ascribed to a difference in response to the reactivation mechanism of the host cell.
