Mechanisms of the electron transfer reaction and the nature of the soluble factor released by irradiation were studied in thymocyte mitochondria. The effects of oxygen and cyanide on the succinic-2-p-iodophenyl-3-p-nitrophenyl- 5-phenylmonotetrazolium chloride (succinic-INT) reductase activity was measured in irradiated thymus homogenates; and succinic dehydrogenase, ubiquinone, and flavin cortents were measured in normal and irradiated (800 rads) thymus preparations. No reduction in the flavin content was found showing that dehydrogenase is not lost from thymus mitochondria following irradiation. Chemical properties of the soluble cofactor are cited. It is concluded that the coupling factor is likely a protein which is labilized from the mitochondria by irradiation. (H.M.G.)
