Air-dried seeds containing 4.92% moisture and sprouts of N. rustica were gamma irradiated and and the nicotine content determined. The percentage of nicotine present varied as a function of the irradiation dose in 10-day seedlings sprouted from irradiated seeds. Values were higher than those of the controls for seeds irradiated at 0.2 to 1 kr, less for the 2-kr irradiated seeds, and higher again at 5 to 10 kr, and a definite peak at 40 kr, but the drop was considerably below that of the controls at 80 kr. In air-dried seeds the nicotine changed with time, decreasing in an irregular pattern to 50% in ten days with a 1-kr dose, while a 2-kr dose caused practically complete degeneration of the compound. When the irradiated seeds were wet, the presence of nicotinic acid resulted in an increased rate of metabolism and formation of nicotine. Depletion of the precursors in the irradiated seedling decreased the rate of nicotine synthesis. The rate of nicotine synthesis is determined by many factors that do not respond equally to irradiation; this explains the waves in the nicotine levels. The important role of nicotinic acid was confirmed in experiments with seeds in dilute nicotine solutions irradiated in vitro and with infiltration of leaves by nicotinic acid. A 10-kr irradiation of the leaves showed no increase in the nicotine. (TTT)
