RADIATION SENSITIVITY OF RIBONUCLEASE AS A PART OF A SYNTHETIC DNA- RIBONUCLEASE COMPLEX (in Russian)

Synthetic DNA-ribonuclease complexes were obtained and characterized. The DNA was obtained from calf thymus, and the RNAase was a commercial preparation, heated to inactivate any impurities. The DNA-RNAase complexes were made in citrate buffer and separated by centrifuging. The enzymic activity of the ribonuclease was determined with RNA as substrate, the acid-soluble RNA being determined spectrophotometrically. RNAase was considerably more inactivated by x irradiation in the complex form than when in the free state. With increasing radiation doses from 0 to 25 or 30 kr, the decreases in activity, 100% being taken as the initial value, were to approximately 60% for RNAase-free or in a mixture with DNA but to about 30% when in the complex form. Free H and OH radicals formed by the irradiation are considered to activate both the surface of the enzyme and the free DNA part. High-energy peroxides and hydroperoxides are formed, and probably the peroxide derivatives oxidized both the DNA part and the RNAase functional groups, further inactivating the enzyme. These reactions could affect the chromosome level. (TTT)
