SPECTROPHOTOMETRIC EVALUATION OF RADIATION DAMAGE TO HEMOGLOBIN (in Russian)

Solutions of rat and pigeon blood hemoglobin, obtained by hemolysis and centrifugation of washed erythrocytes, were x-irradiated for 1 to 120 min at rates of 700 r/min. A hypochromic effect was shown with irradiation increasing from 0 to 80 kr, with the value of epsilon (ratio oi irradiated to unirradiated) for rat oxyhemoglobin decreasing from 100 to 75 at 555 m mu and to 55 at 578 m mu . Pigeon hemoglobin was more radiosensitive than rat. The marked dependence of the change on the dose indicated that the absorption at 578 m mu might be used as a criterion of radiation damage. Irradiation of oxyhemoglobin sharply increased the absorption at 630 m mu , indicating an increase in the methemoglobin. However, values were too variable for this to be used as a criterion. With 56 kr irradiation, considerable change occurred in the protein part of the molecule, with appearance of globin-hemochromogen spectral lines. Oxyhemoglobin irradiated at 2 to 7 kr and incubated at room temperature in 0.019N NaOH was more susceptible to formation of these lines than the unirradiated. Oxygen effects and after-effects were observed in the irradiated solutions. (TTT)
