Polyfunctional radiosensitizers. I. Effects of a nitroxyl biradical on the survival of mammalian cells in vitro

The effect of bis(2,2,6,6-Tetramethyl-1-oxyl-4-piperidinyl)succinate (Ro- 03-6061) on the survival parameters of V-79 Chinese hamster cells, irradiated under hypoxia, has been compared with related monofunctional nitroxyl compounds. The biradical is more effective in reducing the D$sub 0$ value than the three monoradical nitroxyls tested. It also removed the shoulder in asynchronous cell survival experiments, although this effect did not occur in the presence of oxygen. The shoulder for the survival curve for cells irradiated during late S phase was also reduced. Possible mechanisms for its actions are discussed in terms of the bifunctionality of the compound and the potential of such a sensitizer in radiotherapy is emphasized. (auth)
