Daily variation of blood counts with the accumulated dose of BETA radiation was studied after administration of a single dose (0.65 to 5.7 mC) of / sup 32/P to 11 patients with chronic leukemia (nine chronlc myeloid and two chronic lymphatic). White cell count declined to a lower level with larger doses of /sup 321/P but the rate of decline per unlt radiation dose was more with smaller doses. This is explained by the fact that the same amount of radiation dtstributed over a long interval was more effective in lowering white cell count than when applied over a short duration. There was improvement in the erythrocyte count, which was marked more at bigger doses. Maximum depressions of counts of leukemic granulocytes at five days and 35 days or more in leukemic lymphocytes suggested that the turnover time of postmitotic granulocytes was the order of a few days and that in case of postmitotic lymphocytes this was more than a month. These findings also helped in formulating therapy schedules. A dose of 0.05 to 0.1 mu C/g of body weight should be followed by smaller doses at weekly intervals in chronic myeloid leukemia. But a single dose of the order of 0.75 mu C/g may bring about a remission or should be repeated after a month or more in chronic lymphatic leukemia. In one case, remission was attained with a single dose of 0.75 mu C/g body weight for a period of five months. In the other case a repetition of this dosage was necessary by the 35th day. It was concluded that /sup 32/P does not cause general hematopoietic depression nor symptoms of radiation sickness at the doses used. (BBB)
