ABS>To clarify the mechanlsm of the indirect tissue damage by radioinduced lipid changes, lipids of rabbits irradiated with 3000 r were fractionated by silicic acid chromatography and tested for biologic activity on cultures of L cells. Postirradiation incorporation of /sup 32/P-labeled lipids in tissues was also studied. The lipid (lysophosphatide-rich fraction) isolated from irradiated rabbit organs possessed a strong hemolytic action and an ability to inhibit L cell proliferation as well as to accelerate the respiration of mitochondria in the rabbit liver and spleen. It was also shown that they act as to induce a marked swelling of mitochondria, impeding the formation of high- energy phosphate and acting as an uncoupler of oxidative phosphorylation in vivo. The uptake of /sup 32/P-labeled lipids was marked in spleen, bone marrow, and liver of both irradiated and nonirradiate'd groups. The uptake of /sup 32/P- labeled lipids in rabbits given intravenous injections of lipid fractions for 30 consecutive days previously was found to be greatest in pancreas followed by bone marrow, spleen, liver in the male group, whereas it is greatest in spleen, followed by liver and bone marrow in females. It is proposed to lipids liberated and altered by irradiation may act on the bone marrow to depress hematopoiesis. (BBB)
