Development of an ELISA for 1-nitropyrene - A possible marker compound for diesel development exhaust emissions

The exposure to diesel exhaust is a probable carcinogenic risk factor for human beings. Genotoxic substances associated with diesel soot particles such as polycyclic aromatic hydrocarbons (PAHs) and their nitro substituted derivatives (nitro-PAHs) contribute to this carcinogenic potency, possibly. The most abundant nitro-PAH in diesel particulate organic matter is 1-nitropyrene. Among others, both gas chromatography mass spectrometry (GC/MS) and high performance liquid chromatography (HPLC) with fluorescence detection have been used for the determination, both requiring extensive enrichment and clean-up procedures. This talk will discuss the development of an indirect competitive ELISA for 1-nitropyrene including analyte synthesis, protein conjugate preparation, immunization, antiserum characterization, and measurement of real-world samples. Cross-reactivity with both unsubstituted and nitro-PAHs and the effect of organic solvents on the antibody-coating antigen interaction was investigated.
