A novel synthesis of nine- and ten- membered ring lactones mediated by irontricarbonyl

There were few reports on the preparation of physiologically potent ten-membered lactones promoted by transition metals. Of these reactions, however, formation of macrocyclic lactones has so far been restricted to the Stille coupling reaction. In this paper, the authors report that medium ring lactones can be constructed by the intramolecular cyclization of the diene-iron complexes bearing an ester functionality. The iron-mediated intramolecular nucleophilic addition allows for the direct stereocontrol of four stereogenic centers of fused- and bridged bicyclic skeletons. However, increasing the tether length and changing the reaction conditions, for example, intramolecular cyclization of 1 generated medium ring lactones 2-4 in a single step.
