Correlation between ESI-MS, CID, and DNA duplex melting temperature

Electrospray ionization mass spectrometry (ESI-MS) has become an important analytical method for characterizing proteins, pepticles, and small biologically active molecules. One particular class of molecules that are amenable to analysis by ESI-MS are oligonucleotides. Oligonucleotide duplex and small molecule/oligonucleotide duplex noncovalent complexes have recently been studied by ESI-MS. If ESI-MS is to become a useful technique in the analysis and characterization of small molecule/oligonucleotide duplex noncovalent complexes, a comprehensive understanding of the oligonucleotide duplex must be performed by ESI-MS. The goal of this research was to investigate the relationship between the melting temperature ({Delta}T) of an oligonucleotide duplex, ESI-mass spectra and collision induced dissociation (CID).
