Selectivity and binding mode of DNA interacting molecules studied by ESI-MS

An area of interest to a number of scientists are small organic molecules that bind to or interact with DNA, thereby exhibiting antitumor, antiviral, or antibacterial activity. Recently, the noncovalent complex formed between an oligonucleotide duplex and a minor groove binder was observed by electrospray ionization mass spectrometry (ESI-MS). In this report, the authors address two important areas where by ESI-MS may be utilized in the study of minor groove binding molecule/oligonucleotide duplex noncovalent complexes. First, can the binding motif of a minor groove binding molecule be determined by ESI-MS? Second, can differences in the solution binding affinity of minor groove binding molecules to different duplexes be observed in ESI mass spectra?
