A physical map covering 25 cM of human chromosome 8 and including the Langer-Giedion syndrome region

We have developed an integrated physical map of human chromosome 8q23-q24.1 based on STS content analysis of somatic cell hybrids and YAC contigs. Over 50 sequence tagged sites (STSs) were used in construction of the map. These include markers from multiple genetic maps as well as a number of previously unmapped polymorphisms, thus integrating all available short tandem repeat polymorphisms into a single, ordered set based on their positions in the physical map. These markers cover a 25 cM region, with an average spacing of less than 1 cM between markers. The YAC clones used to develop the contig map were identified in two ways: by querying the Genethon Quickmap database for clones connecting STSs mapped to the 8q23-q24.1 region, and by direct screening of the Genethon and other human YAC libraries. All clones identified were tested for STS content to verify their position and extent in the contig. This resulted in detection of a number of gaps in the Genethon contigs caused by the incorporation of chimeric YACs into the paths between STSs or by the lack of resolution of markers in the Genethon genetic maps. The gaps were filled in by library screening with additional STSs to produce a continuous map composed of well-characterized YACs. Using these maps, we have determined the precise locations of the end points of the minimal deletion region involved in the clinical manifestations of Langer-Giedion syndrome. The YAC contigs thus provide the resources for isolation of the causative genes for LGS. Integration of the physical and genetic maps of 8q23-q24.1 will facilitate mapping and cloning of additional disease genes in this portion of the chromosome.
