Solid bovine serum albumin was irradiated with 1- and 2-Mev electrons in the presence of oxygen and adsorbed water. Changes tc solubility and accessibility of disulfide bends and alterations to the protein side chains were examined. The presence of oxygen during irradiation of BSA with a 5% water content had no effect on the change in the Physicochemical properties resulting from one ionization per molecule, i.e., change of shape revealing 25% of the disulfide bonds, but slightly enhanced the production of totally denatured protein with 50% disulfide bonds accessible. Conformation changes in bovine serum albumin in aqueous solution of gamma irradiation

Polarography has been applied to the radiation denaturation of bovine serum albumin in aqueous solution caused by /sup 60/Co ..gamma.. rays. The albumin globules are destroyed and the hydrophilic parts of the polypeptide chain unwind. The concentration of groups capable of electroreduction is thereby increased by more than a factor four. It is found that the conformation is destroyed mainly by OH radicals.

The time-dependent conformational change of bovine serum album (BSA) during Maillard reaction with dextran under controlled dry heating has been studied by small-angle X-ray scattering, fluorescence spectroscopy, dynamic light scattering, and circular dichroism analysis. Through the research on the radii of gyration (R_g), intrinsic fluorescence, and secondary structure, conjugates with dextran coating were found to inhibit BSA aggregation and preserve the secondary structure of native BSA against long-time heat treatment during Maillard reaction. Irradiation of Proteins in the Solid State. II. Chemical Changes Produced in Bovine Serum Albumin

Changes in nine amino acid residues, which make up 73.5% of bovine serum albumin, have been followed after irradiation of this protein in the solid state with 2-Mev electrons in vacuo. The sensitivity to radiation varies by a factor of 2.5. Although carbonyl groups are formed, no evidence of the breaking of the main chain could be detected.
